    Comparative anatomy and evolution of the gastrotrich muscular system

    Gastrotrichs figure prominently in metazoan phylogeny because they share a suite of complex morphological characteristics with several other members of the Bilateria. But their microscopic size, cryptic interstitial habitat, and lack of fossil record have exacerbated the usual barriers to phylogenetic analysis. To arrive at a better understanding of gastrotrich systematics and evolution, cladistic analyses and detailed studies of the muscular system were performed. A fluorescent F-actin stain was applied to whole mounts of 26 species of Gastrotricha to characterize the musculature. Muscle patterns were mapped, their functions inferred, and the direction of evolution hypothesized for several families. The musculature of all gastrotrichs is arranged as a series of circular, helicoidal, and longitudinal bands around the digestive tract. Circular muscles are generally present in splanchnic and somatic positions. Helicoidal muscles in 50--60° angles are present on the pharynx and intestine of most species. Longitudinal muscles are arranged radially around the digestive tract in dorsal, lateral, ventral and ventrolateral positions. Extraordinary muscle orientations are present in several species. In macrodasyidan gastrotrichs, the musculature of Dactylopodola baltica (Dactylopodolidae) is considered to be closest to the ground pattern of the phylum and consists of the following: splanchnic circular muscles on the pharynx and intestine, longitudinal muscles in dorsal, lateral, ventral and ventrolateral positions, pharyngeal and intestinal helicoidal muscles, and somatic circular muscles. Within the Chaetonotida, species of Neodasys and Xenotrichula have the most plesiomorphic muscle topologies. Muscle patterns are similar to macrodasyidans though several muscle orientations have become reduced (splanchnic and somatic circular muscles), are the result of evolutionary modification to existing muscles (incomplete splanchnic and somatic circular muscles, dorsoventral muscles) or evolved independently (the branched Ruckenhautmuskel). This study relied on a phylogenetic perspective to delineate the origin of specific muscle patterns in gastrotrichs and allow for the separation of phyletic heritage from adaptation. Several species from both orders possess muscle patterns that can be regarded as apomorphic and may therefore serve as taxonomic characters. Closer scrutiny of these species may reveal the underlying selective processes that led to the origin and maintenance of novel muscle orientations in gastrotrichs

    Design of a tunable, room temperature, continuous-wave terahertz source and detector using silicon waveguides

    We describe the design of a silicon-based source for radiation in the 0.5-14 THz regime. This new class of devices will permit continuously tunable, milliwatt scale, cw, room temperature operation, a substantial advance over currently available technologies. Our silicon terahertz generator consists of a silicon waveguide for near-infrared radiation, contained within a metal waveguide for terahertz radiation. A nonlinear polymer cladding permits two near-infrared lasers to mix, and through difference-frequency generation produces terahertz output. The small dimensions of the design greatly increase the optical fields, enhancing the nonlinear effect. The design can also be used to detect terahertz radiation

    Convergent synthesis of a steroidal antiestrogen-mitomycin C hybrid using “click” chemistry

    A convergent synthesis of a novel estrogen receptor-targeted drug hybrid was developed based on structures of the potent anti-proliferative mitomycin C and the steroidal anti-estrogen RU 39411. The steroidal antiestrogen was prepared with an azido-triethylene glycoloxy linker while the mitomycin C derivative (porfirimycin) incorporated a complementary 7-N-terminal alkyne. The two components were ligated using the Huisgen [3 + 2] cycloaddition (“click”) reaction.     Paroxysmal nocturnal hemoglobinuria in systemic lupus erythematosus: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Paroxysmal nocturnal hemoglobinuria is an acquired disorder of hemopoiesis and is characterized by recurrent episodes of intravascular hemolysis due to an increased sensitivity to complement-mediated hemolysis. Systemic lupus erythematosus with paroxysmal nocturnal hemoglobinuria is very rare. We report a case of paroxysmal nocturnal hemoglobinuria that developed in a patient with systemic lupus erythematosus and lupus nephritis.</p> <p>Case presentation</p> <p>A 29-year-old Mongolian woman had systemic lupus erythematosus, which manifested only as skin lesions when she was 12 years old. She had leg edema and proteinuria when she was 23 years old, and a renal biopsy revealed lupus nephritis (World Health Organization type IV). She had been treated with steroids and immunosuppressant therapy. At 29, she had headaches, nausea, general fatigue, and severe pancytopenia and was admitted to our hospital. A laboratory evaluation showed hemolytic anemia. Further examination showed a neutrophil alkaline phosphatase score of 46 points, a CD55 value of 18%, and a CD59 value of 78.6%. The results of Ham test and sugar water tests were positive. The constellation of symptoms throughout the clinical course and the laboratory findings suggested paroxysmal nocturnal hemoglobinuria.</p> <p>Conclusions</p> <p>To the best of our knowledge, systemic lupus erythematosus with paroxysmal nocturnal hemoglobinuria is very rare. Clinicians should be aware of the association between autoimmune and hematological diseases.</p

    Rare B decays and Tevatron top-pair asymmetry

    The recent Tevatron result on the top quark forward-backward asymmetry, which deviates from its standard model prediction by 3.4σ\sigma, has prompted many authors to build new models to account for this anomaly. Among the various proposals, we find that those mechanisms which produce ttˉt\bar t via tt- or uu-channel can have a strong correlation to the rare B decays. We demonstrate this link by studying a model with a new charged gauge boson, WW'. In terms of the current measurements on BπKB\to \pi K decays, we conclude that the branching ratio for BπKˉ0B^-\to \pi^- \bar K^0 is affected most by the new effects. Furthermore, using the world average branching ratio for the exclusive B decays at 2σ2\sigma level, we discuss the allowed values for the new parameters. Finally, we point out that the influence of the new physics effects on the direct CP asymmetry in B decays is insignificant.Comment: 15 page, 6 figures, typos corrected and references added, final version to appear journa

    Feasibility of using quadriceps-strengthening exercise to improve pain and sleep in a severely demented elder with osteoarthritis – a case report

    BACKGROUND: Osteoarthritis (OA) of the knee, which is prevalent among older adults in nursing homes, causes significant pain and suffering, including disturbance of nocturnal sleep. One nonpharmacologic treatment option is quadriceps-strengthening exercise, however, the feasibility of such a treatment for reducing pain from OA in severely demented elders has not been studied. This report describes our test of the feasibility of such an exercise program, together with its effects on pain and sleep, in a severely demented nursing home resident. CASE PRESENTATION: The subject was an elderly man with severe cognitive impairment (Mini-Mental Status Exam score 4) and knee OA (Kellgren-Lawrence radiographic grade 4). He was enrolled in a 5-week, 10-session standardized progressive-resistance training program to strengthen the quadriceps, and completed all sessions. Pain was assessed with the Western Ontario and MacMaster OA Index (WOMAC) pain subscale, and sleep was assessed by actigraphy. The patient was able to perform the exercises, with a revision to the protocol. However, the WOMAC OA pain subscale proved inadequate for measuring pain in a patient with low cognitive functioning, and therefore the effects on pain were inconclusive. Although his sleep improved after the intervention, the influence of his medications and the amount of daytime sleep on his nighttime sleep need to be considered. CONCLUSIONS: A quadriceps-strengthening exercise program for treating OA of the knee is feasible in severely demented elders, although a better outcome measure is needed for pain

    TRIB2 as a biomarker for diagnosis and progression of melanoma

    Malignant melanoma is the most deadly form of skin cancer. There is a critical need to identify the patients that could be successfully treated by surgery alone and those that require adjuvant treatment. In this study, we demonstrate that the expression of tribbles2 (TRIB2) strongly correlates with both the presence and progression of melanocyte-derived malignancies. We examined the expression of TRIB2 in addition to 12 previously described melanoma biomarkers across three independent full genome microarray studies. TRIB2 expression was consistently and significantly increased in benign nevi and melanoma, and was highest in samples from patients with metastatic melanoma. The expression profiles for the 12 biomarkers were poorly conserved throughout these studies with only TYR, S100B and SPP1 showing consistently elevated expression in metastatic melanoma versus normal skin. Strikingly we confirmed these findings in 20 freshly obtained primary melanoma tissue samples from metastatic lesions where the expression of these biomarkers were evaluated revealing that TRIB2 expression correlated with disease stage and clinical prognosis.     The OMERACT-OARSI Core Domain Set for Measurement in Clinical Trials of Hip and/or Knee Osteoarthritis

    Objective: To update the 1997 OMERACT-OARSI (Outcome Measures in Rheumatology-Osteoarthritis Research Society International) core domain set for clinical trials in hip and/or knee osteoarthritis (OA). Methods: An initial review of the COMET database of core outcome sets (COS) was undertaken to identify all domains reported in previous COS including individuals with hip and/or knee OA. These were presented during 5 patient and health professionals/researcher meetings in 3 continents (Europe, Australasia, North America). A 3-round international Delphi survey was then undertaken among patients, healthcare professionals, researchers, and industry representatives to gain consensus on key domains to be included in a core domain set for hip and/or knee OA. Findings were presented and discussed in small groups at OMERACT 2018, where consensus was obtained in the final plenary. Results: Four previous COS were identified. Using these, and the patient and health professionals/researcher meetings, 50 potential domains formed the Delphi survey. There were 426 individuals from 25 different countries who contributed to the Delphi exercise. OMERACT 2018 delegates (n = 129) voted on candidate domains. Six domains gained agreement as mandatory to be measured and reported in all hip and/or knee OA clinical trials: pain, physical function, quality of life, and patient’s global assessment of the target joint, in addition to the mandated core domain of adverse events including mortality. Joint structure was agreed as mandatory in specific circumstances, i.e., depending on the intervention. Conclusion: The updated core domain set for hip and/or knee OA has been agreed upon. Work will commence to determine which outcome measurement instrument should be recommended to cover each core domain
